I'm using StarPort v4.2 2009-07-02 on XP x64 and creating a local RAM disk.

Can the default file system for this RAM disk be NTFS so when I reboot it's NTFS and not FAT?

Also, can the drive letter mapping be customized and remain persistent across reboots?

Of course, I can use Disk Manager to make it NTFS and the drive letter what I want but upon reboot it gets reset to FAT and the letter changes.

Unfortunately you cannot save NTFS partition upon reboot.
If you need an NTFS partition on an iscsi target, you can create a local imagefile device and set it to "auto-mount" on re-boot.
